AFM Operation in Fluid

While operating in fluid is essentially the same as operating in air for a given imaging mode, there are certain considerations that must be taken into account to ensure that safety requirements are met, samples remain pristine, and the best possible images are obtained. The considerations for each step in the imaging process are described in this section, given in the order they will occur in the imaging workflow:

  1. Fluid Set Up
  2. Align the Laser
  3. Load the Sample and Adjust the SPM Scanner
  4. Locate the Tip
  5. Tune the Cantilever (TappingMode Only)
  6. Focus on the Sample Surface
  7. Check Initial Scan Parameters
  8. Engage and Adjust Scan Parameters
  9. Withdraw and Clean Up
